چکیده

3',5' cydic-AMP (CAMP) will stimulate the rate of tryptophanase synthesis in Esckrichia coli cultures induced with tryptophan. Adding CAMP after the initiation of messenger RNA synthesis was blocked by rifampicin, did not stimulate tryptophanase synthesis. This indicates that cAMP acts at initiation of either transcription or translation and not at the level of chain elongation of either the messenger or the polypeptide chain. XPERIMENTS using actinomycin D to block RNA synthesis suggested that E cAMP affects the translation of the tryptophanase messenger (PASTAN and PERLMAN 1969). On the other hand, when RNA synthesis was blocked by rifampicin, it was suggested that cAMP affects transcription of the tryptophanase messenger (DEL CAMPO, RAMIREZ and CANOVAS 1970). Since experiments with a temperature-sensitive mutant of Escherichia coli that fails to synthesize functional (Y and p-galactosidase or tryptophanase in the absence of cAMP at 43", pointed out the possibility that the synthesis of these three enzymes is affected similarly by cAMP (SIMON and APIRION, manuscript in preparation), we decided to further investigate the site of action oi cAMP in stimulation of tryptophanase synthesis.

Stimulation of Tryptophanase Synthesis in Escherichia coli by Cyclic 3’, S-Adenosine Monophosphate

Cyclic 3’,5’-AMP stimulates the rate of tryptophanase synthesis in Escherichia coli treated with tris(hydroxymethyl)aminomethane and ethylenediaminetetraacetic acid and induced with tryptophan. A maximal effect occurs at 4 X low4 M cyclic 3’,5’-AMP and a half-maximal response at 1.5 X lo-4 M. The action of cyclic 3’,5’-AMP is specific for tryptophanase since the over-all rate of protein and RNA...

Effect of cyclic guanosine 3,5-monophosphate on the synthesis of enzymes sensitive to caatabolite repression in intact cells of Escherichia coli.

Cyclic guanosine 3',5'-monophosphate inhibits the synthesis of beta-galactosidase and tryptophanase in cultures of Escherichia coli growing in minimal media with glucose or glycerol as the carbon source. Cyclic guanosine 3',5'-monophosphate acts at the transcriptional level in the lac operon, it exerts its action at the promoter site of the operon, and requires the presence of functional cyclic...

Regulation of inducible enzyme synthesis in Escherichia coli by cyclic adenosine 3', 5'-monophosphate.

Cyclic adenosine 3’,5’-monophosphate (cyclic AMP) overcomes the repression by glucose of the synthesis of the following inducible enzymes and proteins in Escherichia co2i: P-galactosidase, lac permease, galactokinase, glycerokinase, L-cx-glycerophosphate permease, Enzyme II for fructose of the phosphoenolpyruvate phosphotransferase system, Larabinose permease, tryptophanase, n-serine deaminase,...

Cyclic adenosine 3',5'-monophosphate-mediated hyperinduction of araBAD and lacZYA expression in a crp mutant of Escherichia coli K-12.

A spontaneous lac+ revertant of an adenylate cyclase deletion strain of Escherichia coli K-12 was isolated and characterized. This revertant, designated strain KC20, exhibited a pleiotropic suppression of the adenylate cyclase defect, with the crp locus being the site of the suppressor mutation. Cyclic adenosine 3',5'-monophosphate at an exogenous concentration of 1 mM severely inhibited the gr...
